Background

PINK1 (PTEN-induced putative kinase 1) is a serine/threonine kinase primarily localized to mitochondria that serves as a critical sensor for mitochondrial quality control through the PINK1/Parkin mitophagy pathway; upon mitochondrial damage, PINK1 stabilizes on the outer mitochondrial membrane and phosphorylates both ubiquitin and the E3 ubiquitin ligase Parkin, initiating the selective autophagic clearance of dysfunctional mitochondria, while loss-of-function mutations in the PINK1 gene are linked to autosomal recessive early-onset Parkinson's disease, highlighting its essential role in neuronal survival and protection against neurodegeneration.
